AstraZeneca Plc (NASDAQ:AZN) on Tuesday reported updated Phase 3 SERENA-6 trial results showing that camizestrant combined with a CDK4/6 inhibitor continued to deliver significant progression-free survival benefits in hormone receptor-positive, HER2-negative advanced breast cancer.

Updated SERENA-6 Data Reinforces Progression-Free Survival Benefit
The updated analysis showed the camizestrant combination reduced the risk of disease progression or death by 55% compared with the aromatase inhibitor regimen.
Median progression-free survival reached 16.8 months for patients receiving camizestrant, versus 9.2 months for those who remained on aromatase inhibitor therapy, representing an improvement of 7.6 months.
Trial Shows Durable Benefit Beyond Initial Progression
For the key secondary endpoint of second progression-free survival, the camizestrant combination reduced the risk of second disease progression or death by 37%.
Median second progression-free survival was 25.7 months for the camizestrant arm compared with 19.1 months in the comparator group, indicating that benefits persisted even after subsequent therapies were administered.
The trial also showed patients receiving camizestrant experienced a longer period before requiring more intensive treatments such as chemotherapy or antibody-drug conjugates.
Median chemotherapy- or ADC-free survival was 22.6 months, compared with 18.7 months for the control arm.
ctDNA Clearance and Regulatory Progress
Exploratory analyses highlighted substantial reductions in circulating tumor DNA.
By week eight, patients switched to camizestrant and recorded a median 99% reduction in total ctDNA, with 51% achieving complete ctDNA clearance.
In contrast, patients continuing aromatase inhibitor therapy experienced a median 64% increase in total ctDNA, while only 1.9% achieved total clearance.
Overall survival data remain immature, though results showed a numerical trend favoring the camizestrant combination. The study will continue through the final overall survival analysis.
